Fmoc-L-Amino Acids were first reported by Carpino in 1970. Peptide synthesis and cleavage conditions are milder with Fmoc amino acids than Boc amino acids. This protecting group offers also more flexibility for the modification of the peptide chain and more specificity in the cleavage of the side-chain protecting groups.
The Fmoc group is removed from Fmoc-peptide chain under basic conditions, thus allowing any acid labile resins to be used. The Fmoc group is usually removed using piperidine, which removes the Fmoc group and reacts with the deprotection byproduct, the dibenzofulvene, to prevent undesired side reactions and byproducts. This dibenzofulvene adduct allows the UV monitoring of the deprotection progression to be performed.
The Fmoc group has a limited stability towards tertiary amines such as DIEA, pyridine, this stability depends on base concentration, solvent and temperature.
